The scene was deemed Under Control at 4.23am.
The fire has been deemed suspicious.
Spencer Street is currently blocked off to traffic in both directions, east of Dudley Street.
While eastbound traffic is expected to reopen within the next couple of hours, westbound will remain blocked for the next few hours.
Update 4.05AM
Around 60 firefighters with 12 primary and specialist firefighting appliances, including aerial platforms, are tackling the blaze, which remains contained to the building of origin.
Approximately 150 people have been evacuated from an adjacent high-rise building.
Residents are being allowed to visit their apartments under police escort while alternative accommodation is arranged.
Victoria Police, Ambulance Victoria and the water and gas companies are on the scene.
2.42AM
At 1.52AM, Fire Rescue Victoria (FRV) was called to an incident on Spencer Street at West Melbourne after a caller to Triple Zero (000) reported a building fire.
Firefighters arrived on scene within five minutes to find a warehouse around 30m x 40m fully involved with fire.
The response was escalated and a community warning was issued for smoke.
The scene is not yet Under Control and firefighters remain on scene.
